Theaflavin 3,3'-digallate is a black tea polyphenol that acts as a potent inhibitor of the Zika virus (ZIKV) NS2B-NS3 protease with an IC50 of 2.3 μM. By directly binding to the proteolytic cavity of ZIKV protease, it blocks the cleavage processing of viral polyprotein precursors. Consequently, this inhibition disrupts essential viral maturation processes, resulting in dose-dependent suppression of ZIKV replication and overall viral growth. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
