Biological Activity

TH1760 is a first-in-class, potent, selective, and cell-active inhibitor of NUDT15 (MTH2). By directly binding into the catalytic pocket of NUDT15, TH1760 inhibits its nucleotide hydrolase function and blocks the clearance of active thiopurine metabolites. Consequently, this target inhibition enhances the accumulation of 6-thio-dGTP in nucleic acids, resulting in the potentiation of thiopurine-induced cytotoxicity and cellular sensitization. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
