Biological Activity

TH10785 is an 8-oxoguanine DNA glycosylase 1 (OGG1) activator that interacts with phenylalanine-319 and glycine-42 residues of OGG1 to increase enzyme activity 10-fold. By controlling catalytic activity through its nitrogen base structure, TH10785 generates a β,δ-lyase function and increases cellular OGG1 recruitment and repair of oxidative DNA damage, shifting base excision repair to a PNKP1-dependent, APE1-independent pathway. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
