Tezacaftor (VX-661) is a small-molecule cystic fibrosis transmembrane conductance regulator (CFTR) corrector that specifically targets the mutant F508del-CFTR protein. By facilitating the proper folding and conformational stability of F508del-CFTR, tezacaftor promotes its cellular processing and trafficking from the endoplasmic reticulum to the plasma membrane. Increased cell-surface expression of functional CFTR channels subsequently restores cAMP-mediated chloride ion transport across epithelial cell membranes. [1]
