Tetrazolium Red (2,3,5-triphenyltetrazolium chloride) is a redox-sensitive histochemical dye that serves as an indicator of mitochondrial dehydrogenase activity. In metabolically active cells, intact mitochondrial dehydrogenases reduce the colorless dye into a bright red, water-insoluble formazan precipitate. Conversely, damaged or necrotic tissue with impaired enzymatic activity fails to reduce the reagent and remains unstained, providing clear visual differentiation to delineate and quantify myocardial and cerebral infarct sizes in ischemic injury models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
