Biological Activity

Tetrathiomolybdate (TM) is a copper-chelating agent that targets the cellular copper efflux protein ATP7B. It induces dimerization of the metal-binding domain of ATP7B (WLN4) through a unique sulfur-bridged Mo2S6O2 cluster, expelling copper ions from the domain and inhibiting protein platination. By forming sulfur-bridged copper-molybdenum clusters with metallochaperones, TM blocks metal transfer functions between copper-trafficking proteins and down-regulates copper delivery to metalloenzymes. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
