Biological Activity

Tetrabenazine is a non-competitive inhibitor of the vesicular monoamine transporter (VMAT), exhibiting higher selectivity for VMAT2 over VMAT1. By inhibiting VMAT2, it disrupts the transportation and storage of monoamines such as dopamine, serotonin, and norepinephrine into synaptic vesicles. This inhibition leads to the depletion of presynaptic monoamine stores and reduces excessive dopaminergic neurotransmission, impairing vesicular monoamine accumulation in central nervous system pathways. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
