Tenovin-6 is a small molecule activator of p53 transcriptional activity and an inhibitor of SIRT1, SIRT2, and dihydroorotate dehydrogenase (DHODH). By inhibiting protein deacetylase activities of SIRT1 and SIRT2, Tenovin-6 increases intracellular p53 protein accumulation and enhances its transcriptional activity. This elevated p53 function triggers cell cycle arrest and apoptosis in various cancer cell lines, while inhibiting DHODH suppresses pyrimidine biosynthesis to impair tumor cell growth. [1]
