Biological Activity

Tenovin-3 is a small-molecule inhibitor of SIRT2, a class III histone deacetylase that regulates cellular stress responses and protein stability. By targeting SIRT2 deacetylase activity, Tenovin-3 impairs sirtuin-mediated signaling and leads to elevated levels of the tumor suppressor protein p53 in MCF-7 breast cancer cells. Consequently, this p53 pathway activation promotes downstream cell cycle arrest and apoptosis to suppress cancer cell proliferation. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
