Tenapanor is a potent, orally active small-molecule inhibitor of sodium/hydrogen exchanger isoform 3 (NHE3). Acting locally within the gastrointestinal tract, tenapanor directly blocks apical NHE3-mediated ion transport to reduce intestinal sodium absorption. Furthermore, inhibition of NHE3 by tenapanor modulates epithelial tight junctions, leading to increased transepithelial electrical resistance and significantly reduced passive paracellular phosphate flux across the intestinal mucosa. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
