TCS-OX2-29 is a potent and selective orexin 2 receptor (OX2R) antagonist exhibiting high selectivity over human orexin 1 receptors (OX1R). By inhibiting OX2R signaling, the compound blocks orexin-stimulated intracellular calcium mobilization and downstream G-protein-coupled signaling cascades. This targeted receptor blockade disrupts central orexinergic pathways, leading to altered regulation of sleep-wake cycles, behavioral arousal, and energy homeostasis in neurobiological models. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
