TCN 201 is a selective N-methyl-D-aspartate (NMDA) receptor antagonist targeting GluN1/GluN2A (p with marked selectivity over GluN1/GluN2B (pIC50 < 4.3). It acts as an allosteric inhibitor by antagonizing glycine binding to the GluN1 subunit within the NMDA receptor complex. Consequently, TCN 201 suppresses glutamatergic excitatory neurotransmission, attenuating calcium influx and downstream neuronal signaling cascades involved in synaptic plasticity and excitotoxicity. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
