TC-067533 is a small-molecule inhibitor targeting severe acute respiratory syndrome coronavirus (SARS-CoV) papain-like protease (PLpro) with an IC50 of 8.7 µM. Through noncovalent inhibition of PLpro, TC-067533 suppresses viral polyprotein processing and blunts the enzyme's deubiquitinase and deISGylase activities. Consequently, this target blockade suppresses SARS-CoV replication in host cells while restoring innate immune response pathways previously suppressed by viral deubiquitination mechanisms. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
