TBB (4,5,6,7-tetrabromobenzotriazole) is a cell-permeable, ATP-competitive inhibitor of protein kinase CK2, demonstrating activity against rat liver CK2 (IC50 0.9 μM) and human recombinant CK2. By suppressing CK2-mediated phosphorylation, TBB downregulates downstream survival pathways including AKT/mTOR and NF-κB, leading to cell cycle arrest and induction of apoptosis in human cancer cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
