Talazoparib tosylate is a potent, orally bioavailable PARP1/2 inhibitor that targets PARP1 (IC50 = 0.57 nM). By inhibiting PARP enzymatic activity and efficiently trapping PARP-DNA complexes, it disrupts base excision repair pathways. This disruption leads to the accumulation of un-repaired single-strand breaks, which convert to lethal double-strand DNA breaks, inducing synthetic lethality and apoptosis in homologous recombination-deficient cancer cells.
