Biological Activity

Oveporexton (TAK-861) is a potent, orally bioavailable orexin receptor 2 (OX2R) agonist with an EC50 ofin calcium mobilization assays. Activation of OX2R stimulates intracellular calcium signaling pathways involved in central sleep-wake regulation. In histaminergic neurons of the mouse tuberomammillary nucleus, OX2R activation induces dose-dependent membrane depolarization with an EC50 of, promoting neuronal excitability and wakefulness-promoting networks. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
