Tacaciclib is an orally bioavailable covalent inhibitor of cyclin-dependent kinase 7 (CDK7). By selectively targeting CDK7, Tacaciclib disrupts transcription and cell cycle progression, leading to downregulation of oncogenic drivers such as c-Myc. This suppression of transcriptional machinery triggers p53 activation and induces apoptosis in acute myeloid leukemia cells, demonstrating potent antitumor activity in preclinical models. [1]
