Biological Activity

T-3775440 HCl is a potent, irreversible inhibitor of LSD1 (IC50 = 2.1 nmol/L) displaying high selectivity over monoamine oxidases MAO-A and MAO-B. By blocking LSD1-mediated histone demethylation, the compound alters epigenetic signaling and restores transcriptional activity of target genes. This downstream mechanism inhibits cell proliferation while inducing differentiation and apoptosis in acute myeloid leukemia cell lines.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
