Biological Activity

SY-5609 is an orally active and highly selective inhibitor of cyclin-dependent kinase 7 (CDK7) with sub-nanomolar binding affinity (sub-nM KD). By inhibiting CDK7 in both the CDK-activating kinase (CAK) and TFIIH complexes, SY-5609 suppresses transcription and disrupts cell cycle control. These molecular effects lead to downstream cellular responses, including inhibition of cancer cell proliferation, cell cycle arrest, and induction of apoptosis. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
