Streptozotocin (STZ) is a glucosamine-nitrosourea derivative that acts as a DNA-methylating and alkylating agent. It selectively enters pancreatic beta cells via the GLUT2 glucose transporter, causing DNA strand breaks and overactivation of poly(ADP-ribose) polymerase (PARP). This cascade leads to cellular ATP depletion, induction of autophagy and apoptosis, and cytotoxic loss of insulin-producing pancreatic islet cells, establishing experimental models of diabetes. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
