STO-609 is a selective inhibitor of Ca2+/calmodulin-dependent protein kinase kinase (CaM-KK), blocking recombinant CaM-KKα (Ki 80 ng/ml) and CaM-KKβ (Ki 15 ng/ml). By inhibiting CaM-KK activity and its autophosphorylation, STO-609 suppresses downstream targets including CaMKI, CaMKIV, and AMPK. Blockade of the CaM-KK/AMPK signaling cascade disrupts cell proliferation, metabolic regulation, cellular growth, and autophagy in various mammalian cell types. [1]

- Preheat media/saline to 37°C.
