Biological Activity

SR8278 is a synthetic, competitive antagonist of the nuclear receptor REV-ERBα. By blocking REV-ERB-mediated transcriptional repression, SR8278 alters circadian rhythm regulation and modulates downstream metabolic gene expression. Treatment with SR8278 increases the expression of key hepatic gluconeogenic enzymes, including glucose-6-phosphatase (G6Pase) and phosphoenolpyruvate carboxykinase (PEPCK), thereby influencing glucose metabolism. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
