Splitomicin is a selective small-molecule inhibitor of the NAD(+)-dependent histone deacetylase Sir2p. By specifically inhibiting Sir2p enzyme activity, Splitomicin disrupts Sir2p-mediated transcriptional silencing at telomeric and mating-type loci in yeast. This inhibition leads to increased histone acetylation, alters chromatin structure, and derepresses gene expression within silenced heterochromatic regions, impairing Sir2p-dependent cellular function.
