Biological Activity

SP 600125, negative control is a methylated derivative that serves as an inactive control for the c-Jun N-terminal kinase (JNK) inhibitor SP600125, which inhibits JNK1, JNK2, and JNK3 (Ki 0.19 µM). While active SP600125 suppresses JNK pathway activity, this negative control analog fails to reduce cAMP-stimulated mRNA levels of downstream targets such as Aa-Nat, Period1, and MKP-1, helping rule out off-target effects in JNK signaling research. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
