SKA-31 is a potent potassium channel activator that opens KCa3.1 (EC50 260 nM), KCa2.2 (EC50 1.9 μM), KCa2.1 (EC50 2.9 μM), and KCa2.3 (EC50 2.9 μM). Activation of these calcium-activated potassium channels induces cell membrane hyperpolarization, promoting endothelium-derived hyperpolarizing factor signaling, vascular smooth muscle relaxation, and lowering of arterial blood pressure in physiological models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
