Biological Activity

SJB2-043 is a potent inhibitor of ubiquitin-specific protease 1 (USP1) that targets the USP1/UAF1 deubiquitinase complex. By inhibiting USP1 deubiquitinase activity, it increases monoubiquitination levels of substrate proteins such as FANCD2 and PCNA, disrupting the DNA damage response pathway and homologous recombination repair. This downregulates DNA repair mechanisms, leading to accumulation of DNA damage and induction of apoptosis in sensitive cell models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
