Biological Activity

Sitagliptin phosphate is a potent inhibitor of dipeptidyl peptidase IV (DPP-4). By inhibiting DPP-4 enzymatic activity, it prevents the degradation of endogenous incretin hormones, including glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). The resulting accumulation of active incretins stimulates glucose-dependent insulin secretion from pancreatic beta cells and suppresses glucagon release from alpha cells, thereby modulating cellular glucose homeostasis and metabolic signaling pathways. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
