Biological Activity

Simufilam (PTI-125) is an orally active small-molecule modulator targeting filamin A (FLNA) to regulate the actin cytoskeleton. By binding and reversing an altered FLNA conformation, it disrupts toxic interactions between filamin A and the α7 nicotinic acetylcholine receptor (α7nAChR) or toll-like receptor 4 (TLR4). This action blocks amyloid-beta 42 signaling through α7nAChR, downstream tau hyperphosphorylation, neuroinflammation, and amyloid accumulation. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
