Biological Activity

Simufilam dihydrochloride is an orally active filamin A (FLNA) modulator that preferentially binds an altered conformation of FLNA to restore its native shape. Reversal of FLNA proteopathy disrupts its aberrant linkages with the α7 nicotinic acetylcholine receptor (α7nAChR) and toll-like receptor 4 (TLR4). Consequently, this inhibition prevents amyloid-β42-induced α7nAChR hyperactivation and TLR4 signaling, thereby suppressing tau hyperphosphorylation, neuroinflammation, and synaptic dysfunction. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
