SHP099 Hydrochloride is an orally bioavailable, allosteric inhibitor of SHP2 (IC50 0.071 μM) that shows no activity against SHP1. By binding to an allosteric site formed by the SH2 and protein tyrosine phosphatase domains, it stabilizes SHP2 in an inactive conformation. This block suppresses downstream RAS/ERK signaling and inhibits proliferation in receptor tyrosine kinase-driven cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
