Biological Activity

SHIN1 (RZ-2994) is a potent dual inhibitor of human serine hydroxymethyltransferase 1 (SHMT1, IC50 = 5 nM) and SHMT2 (IC50 = 13 nM). By targeting SHMT1 and SHMT2, SHIN1 disrupts folate-mediated one-carbon metabolism, leading to a progressive reduction in intracellular purine nucleotide levels. This depletion impairs DNA synthesis and cellular proliferation, thereby effectively inhibiting the growth of susceptible cancer cells in vitro.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
