SGC-AAK1-1 is a 3-acylaminoindazole-derived chemical probe that functions as a potent dual inhibitor of AP2-associated kinase 1 (AAK1) and BMP-2-inducible kinase (BMP2K) with narrow kinome reactivity. Through AAK1 inhibition, SGC-AAK1-1 blocks the phosphorylation of endocytic adaptors to impair clathrin-mediated endocytosis of the LRP6 receptor, thereby regulating canonical Wnt/β-catenin signaling dynamics and receptor trafficking.
