SF2312 is a natural phosphonate antibiotic that functions as a highly potent inhibitor of human recombinant ENO1 (IC50 37.9 nM) and ENO2 (IC50 42.5 nM). By binding to the active site of enolase enzymes within the conserved glycolytic pathway, SF2312 inhibits the interconversion of 2-phosphoglycerate to phosphoenolpyruvate, disrupting energy generation and displaying antibacterial activity under anaerobic conditions. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
