Biological Activity

Sennoside A is a naturally occurring anthraquinone glycoside and irritant laxative isolated from Rhei Rhizoma that displays purgative activity in the intestine. It is metabolized by intestinal microbiota into rhein anthrone, which acts on the colon to stimulate peristalsis and inhibit fluid absorption. Mechanistically, sennoside A downregulates aquaporin 3 (AQP3) expression in mucosal epithelial cells, leading to decreased mucosal water reabsorption and promoted fecal excretion. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
