Biological Activity

Senexin B is a potent and selective small-molecule inhibitor targeting CDK8 and CDK19 Mediator kinases. By blocking the Mediator kinase module, it suppresses estrogen-induced transcription, reduces phosphorylated RNA polymerase II recruitment, and inhibits cancer cell proliferation. Furthermore, targeted inhibition of CDK8 and CDK19 modulates downstream signaling through the PI3K/AKT/mTOR pathway, suppressing xenograft tumor growth in vivo. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
