Biological Activity

Selumetinib sulfate is an oral, non-ATP-competitive inhibitor of MEK1 (IC50 of 14 nM) and MEK2. By inhibiting MEK1/2 activity within the MAPK/ERK signaling pathway, it blocks downstream ERK1/2 phosphorylation. This pathway suppression leads to inhibited cell proliferation, induced apoptosis, and robust anti-tumor activity in preclinical cancer models, including HT-29 colon carcinoma xenografts. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
