Seltorexant is a selective orexin-2 receptor (OX2R) antagonist with high affinity for human OX2R (pKi ~8.0). By selectively blocking OX2R signaling in the central nervous system, it inhibits orexinergic hyperarousal pathways. This targeted antagonism suppresses wake-promoting signaling cascades, leading to reduced locomotion and the promotion of non-REM sleep without disrupting normal sleep architecture.
