Biological Activity

Selisistat (EX-527) is a potent and selective SIRT1 inhibitor with an IC50 of, displaying over 200-fold selectivity over SIRT2 and SIRT3. By inhibiting SIRT1 deacetylase activity, EX-527 prevents the deacetylation of non-histone substrates, resulting in hyperacetylation of p53. This inhibition selectively modulates cellular stress responses, cell cycle progression, and apoptosis through the p53 signaling pathway without affecting class I and II HDAC family members. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
