Biological Activity

Selective JAK3 inhibitor 1 is a potent, irreversible Janus kinase 3 (JAK3) inhibitor that displays marked selectivity over JAK1 and JAK2, as well as other kinases bearing an equivalent active-site cysteine, including BMX, EGFR, ITK, and BTK. By selectively targeting JAK3, it disrupts downstream JAK/STAT signaling pathways, leading to the inhibition of cytokine-stimulated immune responses and suppressing T-cell activation and lymphocyte proliferation. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
