Biological Activity

SEC inhibitor KL-1 is a selective inhibitor of the super elongation complex (SEC) that targets the AFF4-CCNT1 interaction with a Ki of. By disrupting the binding between the scaffolding protein AFF4 and P-TEFb, KL-1 impairs the release of RNA polymerase II from promoter-proximal pause sites. This inhibition leads to a reduced rate of processive transcription elongation and downregulates SEC-dependent gene expression. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
