SD-2301 is a potent and selective PROTAC-based STAT3 degrader. By engaging the VHL–cullin 2 E3 ubiquitin ligase complex to induce targeted degradation of STAT3, it effectively inhibits STAT3 signaling pathways. This targeted degradation enhances dendritic cell function and activates anti-tumor immunity, leading to robust suppression of tumor growth and synergistic therapeutic efficacy when combined with anti-PD-L1 immune checkpoint blockade in vivo.
