SC144 is a first-in-class, orally active small-molecule inhibitor of gp130. It induces gp130 phosphorylation at Ser782 and promotes its deglycosylation, which leads to gp130 downregulation and inhibition of the IL-6/gp130 signaling axis. Consequently, SC144 abrogates STAT3 phosphorylation and nuclear translocation, suppressing the expression of STAT3 target genes and inhibiting cancer cell proliferation and tumor growth.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
