SBI-553 is a brain-penetrant, β-arrestin-biased positive allosteric modulator of neurotensin receptor 1 (NTSR1) with an EC50 of. It selectively promotes NTSR1 downstream signaling through β-arrestin recruitment while bypassing canonical G protein activation pathways. This biased modulation attenuates psychostimulant-induced dopaminergic activity and behavioral responses without inducing the hypothermia or motor deficits associated with balanced receptor agonists.
