Biological Activity
SBI-0640756 is a first-in-class small-molecule inhibitor of translation initiation factor eIF4G1 that disrupts the assembly of the eIF4F complex. By preventing the interaction between eIF4G1 and eIF4E, SBI-0640756 inhibits cap-dependent protein synthesis and suppresses downstream AKT and NF-κB signaling pathways. Consequently, this compound attenuates cell growth, impairs cell survival, and suppresses tumor growth in preclinical melanoma models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
