SB-525334 is a potent and selective inhibitor of TGF-β receptor I (ALK5) with an IC50 of 14.3 nM. It demonstrates 4-fold selectivity over ALK4 and shows no inhibition against ALK2, ALK3, or ALK6. By suppressing ALK5 serine/threonine kinase activity, SB-525334 blocks TGF-β-induced phosphorylation of Smad transcription factors, thereby inhibiting downstream Smad-mediated signaling and nuclear gene activation associated with fibrotic responses. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
