Biological Activity

SB 265610 is a potent, competitive, non-peptide antagonist of the human CXCR2 receptor that displaces the ligands [125I]-IL-8 (pIC50 8.41) and [125I]-GROα (pIC50 8.47). By binding to a domain distinct from the agonist binding site, SB 265610 effectively prevents receptor activation, subsequently suppressing downstream intracellular calcium mobilization, proinflammatory signaling, and neutrophil chemotaxis.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
