Biological Activity

SB 204990 is a cell-permeable γ-lactone prodrug of SB-201076 that acts as a potent and selective inhibitor of ATP citrate lyase (ACLY). By suppressing ACLY activity, SB 204990 blocks the conversion of citrate to cytosolic acetyl-CoA, thereby attenuating de novo fatty acid and cholesterol biosynthesis. This metabolic inhibition significantly decreases plasma cholesterol and triglyceride levels in vivo and suppresses the proliferation and survival of tumor cells dependent on aerobic glycolysis. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
