(S)-2-Allylglycine (L-Allylglycine) is an amino acid analog that acts as an inhibitor of glutamic acid decarboxylase (GAD), the key rate-limiting enzyme responsible for gamma-aminobutyric acid (GABA) biosynthesis. By inhibiting GAD activity, it suppresses central GABA synthesis, leading to depletion of inhibitory GABA levels in the brain. This impairment of GABAergic neurotransmission disrupts neural inhibition and promotes central nervous system hyperexcitability, frequently resulting in seizures and neuronal damage in experimental seizure and epilepsy models.
