Biological Activity

RTICBM-189 is a brain-penetrant negative allosteric modulator of the cannabinoid type-1 (CB1) receptor. By modulating CB1 receptor signaling, it regulates intracellular calcium mobilization and cAMP pathway signaling downstream of the receptor. Pharmacological evaluation demonstrates selectivity over more than 50 off-target proteins and high central nervous system exposure, which significantly attenuates the reinstatement of cocaine-seeking behavior in rat models without altering locomotion. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
