RPI-1 is a 2-indolinone Ret tyrosine kinase inhibitor. In human papillary thyroid carcinoma TPC-1 cells harboring the RET/PTC1 rearrangement, RPI-1 inhibits cell proliferation and promotes G2 cell cycle phase accumulation. It abolishes Ret/Ptc1 tyrosine phosphorylation and disrupts its binding to Shc and phospholipase Cγ, abrogating downstream JNK2 and AKT activation pathways to block oncogenic Ret signaling. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
